popup.description1
The aim of the study is to investigate the influence of various epigenome modifiers and regulatory elements in genetic constructs on the efficiency of plant genetic transformation and expression of transferred genes, as well as the effect of genetic transformation and epigenome modifiers on the synthesis of secondary metabolites in plants.
popup.description2
Object – plant in vitro systems capable of synthesizing recombinant proteins and biologically active compounds.
Aim – to investigate molecular, physiological-biochemical, and morphogenetic processes that determine the efficiency of expression of transferred genes in plant systems. This includes cloning and functional evaluation of structural and regulatory sequences, development and analysis of genetic vectors, as well as studying the stability of transgene expression, morphogenesis, and the regeneration potential of various plant objects. Particular attention is paid to the use of transformed plants as sources of biologically active compounds and as platforms for the “green” synthesis of metal nanoparticles.
Research methods: in vitro culture, Agrobacterium-mediated transformation, vector construction, Golden Gate molecular cloning, PCR/RT-PCR, FTIR spectroscopy, transmission electron microscopy, surface plasmon resonance, and analysis of data from the online database QuinoaDB.
During the study, a TVCV-based vector for expression of the infα2b gene was created, providing a higher level of interferon accumulation compared with the PVX system. Fusion with calreticulin and a His₆ tag increased the efficiency of protein secretion and purification. A vector system with two reporter genes for soybean transformation using A. tumefaciens GV3101 was developed, and regeneration was optimized. Stable expression of colicin in transgenic kale lines with a heterosis effect in the T1 generation was demonstrated. A high potential of the Lemnaceae family for recombinant protein expression was established. Genes of Amaranthus hypochondriacus associated with organogenesis were identified, and a positive effect of polyethylene glycol on tomato regeneration was confirmed. The prospects of “green” synthesis of silver and gold nanoparticles from extracts of transformed roots of Artemisia annua were demonstrated.
