popup.description1
To determine the impact of certain pre-treatment approaches on the efficiency of cryopreservation, hypothermic storage and experimental transplantation of cell structures with different levels of organization.
popup.description2
The study focused on mesenchymal stem/stromal cells (MSCs) and the paracrine factors (PFs) they produce. The research aim was to investigate the effects of MSC preconditioning by encapsulation in alginate microspheres on selected metabolic parameters, mitochondrial membrane potential, and resistance to oxidative damage at different levels of cellular organization; and to evaluate the effectiveness of adding MSC-secreted PFs to a hypothermic preservation solution for isolated rat livers. The results demonstrated that, as early as the first day after encapsulation in alginate microspheres, MSCs exhibited reduced metabolic activity, impaired mitochondrial function, and decreased basal and stimulated production of reactive oxygen species compared with cells cultured in a monolayer. Furthermore, cell cycle arrest at the G1 phase was observed in all cells 48 hours after encapsulation. Taken together, the decreased metabolic activity and reversible cell cycle arrest indicate that MSCs enter a quiescent state following encapsulation in alginate microspheres. It was established that PFs, present in conditioned media and added to the preservation solution for isolated livers, reduced oxidative damage to the organ, preventing its further increase following normothermic reperfusion. The presence of PFs in the preservation solution positively affected mitochondrial respiratory activity by preserving mitochondrial barrier function and improving the coupling of respiration and phosphorylation, resulting in increased hepatic ATP content at all stages of the experiment. Supplementation of preservation solution with PFs also substantially affected the clearance of injury markers from the liver during normothermic reperfusion, partially prevented the deterioration of secretory function, and significantly improved the organ's morphological condition, which remained close to that of the control group.
