The goal of this project is investigation of the processes which will be take place during the two human gene delivery - insulin and main protein component of high-density lipoproteins apolipoprotein A1(Аpо АI) - on model systems in vitro and in experimental animal organisms. We expect that reproduction of insulin synthesis in organism (wherewith creation of insulin producer cells in the liver) and high-density lipoproteins higher level synthesis, that is one of the key constituent for vessels maintenance, will enable to create scientific and technological prerequisites for medical treatment of Type 1 diabetes and recovery of the organism from vascular lesions, which are complications of main pathology. For 4 month period under review in accordance with plan basicrecombinant vectors for expression of human apolipoprotein A1 under strong hybrid eucariotic promoter regulation and molecular constructions for gene delivery of reporter genes and target preproinsulin gene, which is the key in carbohydrate metabolism regulation and energy homeostasis maintenance, have been constructed. The initial descriptionsof lipid metabolism state of rabbits, which will be used for the making of target model pathology, have been obtained.
