Karazin popup.description2 Object of study - male Wistar rats 3- and 24-months of age, which kept on a standard vivarium diet, and 3-month average level of animal locomotive activity that were exposed to chronic neurogenic stress and N-acetylcysteine. In addition, the objects of study were: isolated hepatocytes and isolated brain structures, the neocortex and hippocampus of 3- and 24-month-old Wistar rats. Purpose - to clarify the role of neutral sphingomyelinase in deviation of the functional state of cells and tissues during aging and in conditions of stress and in the induction of premature aging. Research methods. The paper used spectrophotometric, radioisotope, electrophysiological, chromatographic methods and statistical analysis. The role of ceramides, which are formed in the cells upon activation key ways of accumulation and degradation in hormonal signaling deviation and physiological condition of the animals in the old age and under simulated stress. The medical sphingolipids metabolism modulators (such as N-acetyl-cystein) and specific sphingolipids de novo synthesis reaction and ceramide metabolism inhibitor (miriocin) were used. Particular attention was paid to research the role of neutral sphingomyelinase functional status changes in biochemical composition of organs and tissues in old age, or under the influence of artificially induced stress, which accelerates the aging process. In addition, investigation embraced changes in the sphingomyelin cycle and cardiolipin metabolism in the hippocampus, neocortex, and liver and cognitive function during normalizing metabolism of sphingolipids using neutral sphingomyelinase inhibition with N-acetylcystein. The work also aims to clarify the characteristics of bioelectrical brain activity of rats with high and medium mobility levels under conditions of chronic neurogenic stress and emotional effects of chronic stress on sphingo and - glyceroolypid content in certain brain structures. In addition, using functional reaction inhibitor of neutral sphingomyelinase - N-acetylcystein the contribution of this enzyme in stress-induced changes in behavior and physiological properties of rat brain was studied.
