Investigation object is the process of obtaining a highly active virus-safe preparation of factor VIII blood clotting with the use affinity chromatography method on macroporous silica sorbents. The aim of the work - development of a obtaining method of highly active virus-safe preparation of blood clotting factor VIII with the use of newly synthesized biospecific silica sorbents with immobilized active dyes and іnvestigation the obtained preparations properties. Research methods: chemical synthesis of sorbents; pre-fractionation of blood plasma proteins; dialysis; gel and ultrafiltration; spectrophotometric; electrophoretic; affinity and ion-exchange chromatography; coagulogic and methods using hydrolysis of specific chromogenic substrates; antiviral treatment with solvent-detergent and thiocyanate methods; immunoenzyme; statistical. As a result of the work were obtained the following scientific and practical results: were synthesized and tested in the experiment a series of amino and hydroxypropyl silica derivatives with immobilized dye ligands and was demonstrated their suitability for purification of blood clotting factor VIII; were investigated the optimal conditions for the use of synthesized sorbents (stability at different pH, ionic strength, in the presence of polar or non-polar solvents, chelators, etc.); conditions for sorption and desorption of factor VIII and non-target proteins for effective purification of the protein that was investigated. Degree of implementation: acts of implementation, reports at international scientific forums, publications in the scientific literature. Industry of application: blood centers, chemical-pharmaceutical enterprises, research organizations. Effectiveness. The implementation of the results of the planned studies will improve the previously established technologies for obtaining diagnostic and therapeutic preparations of blood clotting factor VIII from donor plasma and cryoprecipitate using pre-PEG and salt fractionation, biospecific chromatography on macroporous sorbents. This will increase the efficiency of the processes, reduce the time spent on technology implementation, minimize the loss of activity of the investigated protein in the purification process. In general, for the reporting period were published 20 scientific works, including 2 articles, 17 abstracts, 7 of them abroad, 1 newsletter.
