The aim: to assess the effect of previous chemoradiotherapy on the character of radiation- and chemical-induced cytogenetic effects during radiation therapy in vivo and ex vivo depending on the mutagenic factor and tumor localizations. Research methods and techniques: cytogenetic analysis of human blood cultured lymphocytes, statistical methods of data processing, microscopes Axioskop, Micros MC300X, Olympus CX41 and Olympus, system ASIS. Theoretical and practical results: the influence of previous treatment on the accumulation of radiation-induced aberrations in cancer patients depending on the tumor localizations was found. The increase of radiation-induced aberrations from beginning to end of treatment in patients with lung cancer with previous chemo- or radiation therapy, and less pronounced changes in these parameters in patients with head and neck tumors were demonstrated. Against the background of the absence of a pronounced effect of chemotherapeutic treatment on the frequency of radiation-nonspecific chromatid type damages a dependence of these parameters on the tumor localizations was observed. In ex vivo model experiments the regularities of the yield and distribution of chromosome aberrations in irradiated patients’ lymphocytes against the background of chemotherapeutic drugs effect were found. Novelty: the quantitative yield and quality range of cytogenetic damage in blood lymphocytes of patients with lung cancer and head and neck cancer before and during radiation therapy in the presence of breaks in exposure or in the presence of previous chemoradiation therapy were identified.
