At the in vivo stage, an animal model of alimentary vitamin D deficiency in rats was developed and verified based on the level of 25OHD in the serum. Animals with different vitamin D status were immunized with the Pfizer-BioNTech mRNA vaccine (10 μg/animal). It was shown that after immunization, the level of 25OHD was decreased in the control and D-deficient groups compared with non-immunized rats of these groups, which may indicate disturbances of metabolism and/or transport of vitamin D3. No statistically significant difference in the level of IgG to the SARS-CoV-2 spike protein depending on the vitamin D status of rats was found, however, a tendency to a decrease in their levels after the development of vitamin D deficiency was observed. When assessing the functioning of the B- (by TGFβ+) and T-cell (by CD46+) immunity in the spleen, multidirectional changes in the cell number were found in non-immunized and immunized rats with different vitamin D status. It should be noted that the restoration of adequate vitamin D status led to a significant decrease in the number of both types of cells that are immunosuppressive, which, in turn, correlated with the highest levels of IgG to the SARS-CoV-2 spike protein. Impairments in the functioning of the vitamin D-auto/paracrine system at the mRNA and protein levels of VDR, VDBP, CYP27B1 and CYP24A1 in the kidneys and lungs of rats with different vitamin D status, immunized with the Pfizer-BioNTech mRNA vaccine were detected. New data were obtained regarding the relationship between the intensity of the immune response of animals vaccinated with the Pfizer-BioNTech, the level of the vitamin D3 bioavailability and the state of the vitamin D-auto/paracrine system in the target organs of animals, which altogether will allow to substantiate the pharmacological effectiveness of the therapeutic use of vitamin D3 supplementation for SARS-CoV-2 infection treatment and on the background of vaccination against COVID-19.
