Bogatsky National Academy of Sciences of Ukraine popup.description2  Objects of study: serrathiopeptidase Serratia E15, carbopol, stimulator of tissue regeneration. The goal is to study the physico-chemical and biochemical properties of a wound-healing and antimicrobial gel with the proteolytic enzyme serratiopeptidase, an antiseptic agent and a tissue regeneration stimulator based on carbopol, and to carry out its medico-biological verification. The main physicochemical and biochemical physicochemical features of serratiopeptidase immobilized in a carbomer gel are determined. It is shown that immobilization contributes to increasing the operational stability of the enzyme in the processes of heat and pH inactivation (40-60 oC; pH 4-8). The results of kinetic studies indicate certain diffusion limitations due to the gradual dissolution of carbopol and a decrease in the rate of formation of the enzyme-substrate complex for the immobilized enzyme, since the KM for the immobilized enzyme is higher by 19.65-28.45%. Conducting a set of studies on the testing of a gel for the treatment of the same type of burns on the skin of IIb rats showed that the use of a gel with serratiopeptidase and dexpanthenol, in contrast to the treatment with "Iruxol" ointment, prevented the development of microbial invasive infection, contributed to faster and more effective cleaning, stimulation of wound regeneration, shortening the healing period by 2-4 days, avoiding the formation of rough scars and restoration of the hair cover of the skin of experimental animals.
