Its expression level was analyzed by Western blot and confocal microscopy. It was found that PC12/GFP-Ruk/CIN85 cells were characterized by the ability to form clusters and to clone growth. The ability to differentiate by neuronal type was detected to be completely blocked in PC12/GFP-Ruk/CIN85 cells, whereas the wild-type cells formed spines in a medium with low serum content after NGF addition, which were similar to the neurons of the sympathetic nervous system. In addition, overexpression of Ruk/CIN85 led to constitutive activation of proteinkinase B (Akt) and increased activation of ERK1/2 after NGF action. A high expression level of Id1 and low expression level of MATH2/NEUROD6 were found in EGFP-Ruk-overexpressing cells compared to control cells.
