The aim of the work is to establish the main parameters of spheroid formation and morphofunctional profile for heterogeneous spheroids from stem/progenitor cells and to determine their osmotic characteristics in dimethylsulfoxide (DMSO) cryoprotectant solutions. Theoretically calculated filtration coefficients Lp and permeability coefficients kp for water and DMSO for a number of bioobjects (cells, SF, cells depending on localization in SF). It was found that for cells of the L929 line, the coefficients Lp and kp are the largest for cells in suspension and decrease for cells in the composition of a spheroid with an increase in the depth of their location. Optimal modes of cryopreservation of SF from L929 cells were calculated, depending on the terms of cultivation. The necessary conditions for the formation of spheroids from MSCs have been determined and the following parameters have been established: surface-to-volume ratio, osmotically inactive volume, Lp and kp coefficients for water molecules and DMSO cryoprotectant, values of the activation energy of their penetration. According to the calculations of physical and mathematical modeling, the range of optimal cooling rates of spheroids obtained from MSC is determined to be 0.75 ÷ 1 deg/min, the recommended concentration of DMSO is 7.5% (1 M).
