Homologous models of human methionyl- and leucyl-tRNA synthetase enzymes were built. A significant difference in the structure of active sites between human and Mycobacterium tuberculosis enzymes was revealed, which is important for the development of new inhibitors selective for pathogens. Genetic engineering constructs were created for obtaining the recombinant human LeuRS and MetRS proteins. New effective inhibitors of the FGFR1 protein kinase were developed among benzimidazole derivatives. Among the compounds studied in vitro, 4 inhibitors were found to be active at nanomolar concentrations, the most effective of which inhibits FGFR1 with IC50 = 150 μM. An analysis of the “structure-activity” relationship of FGFR1 inhibitors was performed and structural elements that affect the biological activity were identified. When studying a series of compounds in the enzymatic DNA relaxation test system in vitro, acridine and phenazine derivatives were identified that inhibit the topoisomerase I enzyme at micromolar concentrations (80-100 μM). The features of the primary and secondary structure of the minimal PBS subdomain (primer binding site) of the genomic RNA of different subtypes of the HIV-1 virus were studied, a phylogenetic analysis and prediction of the secondary structure of this subdomain were performed for more than 400 genomic sequences of the group M. The corresponding sections of the database were created. Restructurings within PBS were analyzed. The results obtained may become the basis for the creation of new drugs for the treatment of oncological, bacterial and viral diseases.
