Highly purified (purity >95%) recombinant enzymes of leucyl- and methionyl-tRNA synthetase of the tuberculosis pathogen Mycobacterium tuberculosis were obtained. Inhibitors of these enzymes active at micromolar concentrations were found, including the multitarget inhibitors that simultaneously inhibit the activity of both enzymes. A number of new inhibitors of the protein kinase FGFR1 based on indazole derivatives were identified, including compounds effective at nanomolar concentrations (IC50 100-330 nM). A series of new cyanine derivatives, the inhibitors of topoisomerase I, active in vitro at low micro- and nanomolar concentrations (3 compounds with IC50 500-800 nM) were obtained. Among them, compounds with antiproliferative activity with EC50 values in the range of 500-900 nM were identified. The structure-activity relationship of enzyme inhibitors was analyzed. Computer models of ligand-target complexes for FGFR1 and topoisomerase I were built. Information was obtained on the structural and functional features of specific structures of the genome of HIV-1 and SIV viruses, in particular, structures covering the PBS domain and hairpins in genomic RNAs. A corresponding section was created in the CESSHIV-1 database. The results may become the basis for the development of new drugs for the treatment of oncological, bacterial and viral diseases.
