Pharmacophore models of MetRS and LeuRS enzymes of the tuberculosis pathogen M. tuberculosis were built. As a result of pharmacophore screening of the library of compounds and biological testing of selected substances in vitro, 5 dual inhibitors of both enzymes, active in micromolar concentrations, were found. Models of binding of FGFR1 protein kinase inhibitors to the ATP-acceptor site of the enzyme were developed, which allowed to explain the data on the dependence of the inhibitory activity of indazole compounds on their structure. New isomeric derivatives of monomethinecyanines containing amino substituents at C-4 position of the quinoline fragment and inhibiting topoisomerase I in vitro at micromolar concentrations were obtained. Biophysical studies indicated the efficient binding of cyanines to DNA. The binding constants of active TopoI inhibitors of the cyanine class to DNA have been determined, the values of which are in the range of (0.7-4.3)×10exp6 M-1, confirming high affinity of ligands. The “structure-biological activity” relationship for the inhibitors of all the enzymes studied was analyzed. The secondary structure was predicted and the functionality of the domain in the genome of HIV-1 and SIV viruses that controls the programmed frameshift during translation of the gag gene was studied for several thousand virus isolates,. The results of this work may become the basis for the development of new drugs for the treatment of oncological, bacterial and viral diseases.
