The objective of the investigations is development of principles of wheat registration using molecular-genetic markers. The following tasks were posed: development of the procedure of electrophoresis of gliadins, the compiling of the catalogue of alleles for the new procedure, creation of the database of genetic formulas of wheat varieties and lines with respect to storage protein loci, development of principles of registration using DNA markers. Methods of investigation are acid polyacrylamide gel electrophoresis of grain proteins, SDS electrophoresis of grain proteins, methods of genetic analysis, PCR analysis. Conditions of SDS electrophoresis of high-molecular-weight glutenin subunits and gliadins encoded by homoeologous group 1 chromosomes, methods of sample preparation were optimized. The simple and effective method of acid electrophoresis of prolamins was developed. The project of the catalogue of alleles of the gliadin loci Gli-A1 , Gli-B1 and Gli-D1 for the acid PAG procedure developed. Electrophoretic analysis of seeds of wheat varieties was made and their formulas with respect to gliadin alleles were determined. Using information from literature sources and our own results, the database of genetic formulas of wheat varieties with respect to storage proteins was compiled. Genetic formulas of about 500 varieties were written. The catalogue includes all published variants of genetic formulas of the same variety. On the basis of results of RAPD and SSRP analysis of wheat varieties of different ecological-geographical origin, methods of compiling genetic formulas of varieties and their registration were developed.
