Schemes of affinity purification of adapter protein Ruk recombinant Glu-tagged isoforms expressed in baculoviral and mammalian expression systems were developed It was established that affinity purified preparations of Ruk protein possess the ability to interact with regulatory subunit of PI 3-kinase and to inhibit in such way its catalytic activity, bind to and hydrolize DNA. It was revealed that the interaction with PI 3-kinase is mediated by SH3 domain of regulatory subunit and Pro-rich region of Ruk, and interaction with nuclease - by coiled-coil region of Ruk.
