We isolated a collection of mutants defective in pexophagy (pdg mutants) and developed an allyl alcohol-based selection scheme for the cloning of PDG genes by functional complementation. The aims of our research work were to study of PDG proteins by intracellular localisation detection and to look for other proteins, interacting with them (2) detection of pupatetive vacuole transport componetns desrupted in each mutants. Among cloned genes, PDG3 (UGT51B) is predicted to encode a sterol glucosyltransferase, whose product is ergosterol glucoside (SG). We observed that a P. pastoris pdg3 UV-induced and constructed deletion mutant are defective in micro- and macropexophagy, as well as in the glucose-induced degradation of cytosolic enzymes. The micropexophagic process in the pdg3 mutant is blocked at the stage of homotypic vacuolar fusion. At the same time, this mutant exhibits normal vacuole morphology, and the process of nitrogen starvation-triggered general (bulk) autophagy was only partially affected. pdg3 mutants were more sensitive to elevated pH and high extracellular ethanol concentrations relative to the wild-type strain.
