Object: sceletogenic cells culture on the matrices in the intervertebral disc repair. The experimental study in vitro and in vivo (90 newborn rats, 146 rats different ages) of the connective tissue cells extracted from young and old rats tissues by different methods and then cultivated in the different culture types. The repair process was evaluated at the introduction into damaged zone of intervertebral disc of the cultivated cells. It was established that the consecutive use of the ferment complex during 12 hours only provided the extraction cells from intervertebral disc and articular cartilage of young rats free of remains of intracellular matrix. 24 hours was required for complete separation adult rat tissue. It was proved the remains of intracellular matrix and cell clusters had the negative influence on the proliferating activity of the cells at their cultivation. High density culture provided the differentiation of the intervertebral disc cells towards cartilage differon and cartilage phenotype safe. It was proved that cultivated cells of intervertebral disc of 12 months old rats was characterized lower proliferation and biosynthesis compared with cell culture 1 months old rats. Cultivation of chondroblasts, annulus fibrosis cells and fibroblasts on the collagen matrices was accompanied stepwise filling of the matrix. At the cultivation stage cells kept their phenotype, their ultrastructural organization indicated substantial metabolic potential. Intervertebral disc injury with nucleus pulposus destruction resulted in the animals both ages (3 and 12 months) destructive disturbances intervertebral disc more expressed in the old rats. Transplantation into injured zone of the intervertebral disc of autologic cells of cartilage differon cultivated on the collagen matrix was accompanied by repair process activation and formation large areas of chondroid, restoration of disc high. Repair process intervertebral disc at cell transplantation did not proved restoration disc structure.
