However many genes are not functionally characterized yet. An important part of this problem is studying of paralogous genes, that are produced by gene duplication and in contrast to ortologous genes not always have identical functions. The aim of this project was identification of binding differences of two members of adaptor protein family intersectins (ITSN), encoded by paralogous genes and involved in endocytosis, cytoskeletal rearrangements and cell signaling. For the first time we showed an interaction of ITSN2 with endocytic protein dynamin 1, Ras guanine nucleotide exchange factor Sos1, ubiquitin ligase c-Cbl and semaphorin 6A. The results obtained indicate that ITSN1 and ITSN2 have similar functions and differ by affinity and specificity of interactions with protein partners. Differences in expression levels of ITSN1 isoforms in the tissues of embryo with trisomy of chromosome 21 and in glial tumors were detected.
