Rapid increase of phospholipase D activity was found in cytokinin-treated plants comparing to untreated ones. The results indicate participation of phospholipase D in early events of cytokinin signaling. Pretreatment of plants with calcium channel blocker verapamil, calcium chelator EGTA, and phosphatidylinositol bisphosphate chelator neomycin dramatically reduced cytokinin-dependent phospholipase D activity. Sensitivity of phosphatidylbutanol accumulation to phospholipase D cofactors - calcium and phosphatidylinositol bisphosphate - suggests a role of different phospholipase D isoenzymes in cytokinin signaling.
