The purpose of research: design vectors and selection of new yeast producers of intracellular and secretory rhARGI and optimization of fermentation. Methods: genetic engineering, molecular genetics, microbiology, biochemistry. Using the methods of metabolic engineering the stable overproducers of recombinant arginase in the two yeasts strains Saccharomyces cerevisiae and Hansenula polymorpha were designed. Target product was obtained as His- and GST-labeled secretory (fused with the leader sequence for secretion MFa, which is frequently used in yeast expression system for production target protein in the culture medium) and intracellular forms of rhARGI. For the expression of relevant chimeric genes of rhARGI in H. polymorpha were used constituent (GAP) and inducible (gene of formate dehydrogenase, FMD) promotors. Between the open reading frame of rhARGI and sequences for purification the sequences for proteolytic cleavage by commercially available enzyme enterokinase have been introduced for production of the final product , an identical native hARGI.
