The aim of the research is primers design for investigation of variability of bovine leukemia virus circulating in farms in Ukraine by means of polymerase chain reaction; the determination of phylogenetic relationship of bovine leukemia virus isolates circulating in different regions of Ukraine; primers design for avian leukosis virus subgroup J detection by means of polymerase chain reaction. The methods of investigations are molecular genetic and bioinformatic. The four primer pairs for amplification of gene pol most variable fragments of bovine leukemia virus provirus DNA are theoretically constructed and synthesized. The temperature and temporal rates of standard polymerase chain reaction with use of design primer pairs are worked out. The bank of BLV isolates circulating in Ukraine is created. The early designed molecular genetic test-system "BLV-provirus DNA-тест" for bovine leukemia diagnostic is improved and registered. The sequences of gene env of BLV provirus DNA from different geographic regions of Ukraine are sequenced. Their primary structures are compared with respective BLV provirus DNA fragments from EMBL, GenBank. It's established that BLV circulating in Ukraine is membership of European-Asian subgroup. The genetic propinquity of BLV and syncytial virus, Jembrana disease virus and BIV is established. Primers for ALV-J detection by means of PCR are constructed, and analysis protocol is developed. The received results can be used in veterinary medicine and laboratory diagnostics. The efficiency of the results application is 8 grv for 1 grv of costs.
