A purpose of work is creation of method of molecular diagnostics of basic viruses of potato by the method of PCR and Real-time PCR. Development of method of making healthy of potato is with application of the designed microgravity. It was design and synthesis of molecular primers for PCR of diagnosticums of potato viruses. Diagnostic test-systems is created for detection of basic viral infections by the method of PCR in a planting-stock and during a vegetation. The terms of PCR and Real-time PCR are optimized. It was determine partial sequences of nucleotide of genes of capsid proteins of the Ukrainian isolates of X-, Y-, M- and S-viruses of potato, and phyllogenetic analysis are carried out. Diagnostic test-systems is created for quantitative determination of the Ukrainian isolates of X- Y- M- and S-viruses of potato. The biotechnology of constructing of plazmid standard, which allows to determine the quantitative loading of genomes of the Ukrainian isolates of PVX is developed, PVY, PVM and PVS, by the method of Real-time PCR. The method of making healthy of plants of potato is developed from a viral infection by cultivation of donor-type plants at the terms of the designed microgravity, introduction of explants of donor-type plants, in the culture of in vitro, receipt of regenerantiv and clonal micropropagation.
