After in vitro expansion cells demonstrated immunophenotype CD29+, CD44+, CD73+, CD90+, CD105+ and capacity to multilineage differentiation, that corresponds to specific for mesenchymal stem cell (MSC) properties. During two-dimensional co-culture of BM MSC and macroporous spongy matrices based on polymeric agarose or alginate cryogels (MACS) there was no appearance of the decrease in proliferative potential of cells. MSC weakly adhere to agarose or alginate MACS, but adhesive properties of MACS could be improved by the modification of cryogels by the addition of type A gelatin. The chemical modification of MACS, prepared by the coupling of "anchor" nanoparticles of type A gelatin to the pore surfaces of macroporous cryogels was more effective than simple incorporation of gelatin. It was obtained that one of the most important factors for the optimal realization of MSC proliferative potential during three-dimensional culture within MACS is the cell seeding step.
