Research object: physico-mathematical modeling of processes, being present at low-temperature preservation of cell suspensions, experimental determination of transport characteristics of cell membranes, geometric and mechanic parameters, cryoresistance under different cooling regimens of human erythrocytes, rat hepatocytes, kidneys of golden hamster, SPEV cells, testes cells, mice ovaries and embryos, Saccharomyces cerevisiae yeast cells. Methods of physico-mathematical modeling, thermodynamics of non-reversible processes, classical theory of phase transformations, hydrodynamics of binding solutions, light low-angle scattering, ion-selective electrometry, light microscopy, tenzodilatometry, viscosimetry, osmometry, thermography, program cooling as well as common methods of cell functional state estimation. Derived physico-mathematical model, describing extra- and intracellular solutions state in cell suspension during its crystallization, enabling to obtain quantitative estimation for optimal regimen of its cooling taking into account the cell transport characteristics. Analysis of crystallization peculiarities of cryoprotectants' aqueous solutions was carried out. It is shown that crystallization of cryoprotectants' aqueous solutions cannot be described with phasal diagrams of eutectic type. There were formulated principals of unbalanced diagrams which response phasal transitions during regimes really using in practical cryobiology. Novelty: the built theory using calculation it is possible to forecast cooling optimal regimes and composition of cryoprotective medium for cells with different functional specialization which accelerate the working-out of different origin cell suspensions low-temperature preservation and decrease their value.
