Development of effective methods of arresting secondary immune deficient states of viral genesis. popup.stage_title Head Tsutsayeva Alla A., Registration Date 06-04-2011 Organization Institute for Problems of Cryobiology and Cryomedicine of the National Academy of Sciences of the Ukraine popup.description2 The research objects were plasma proteins, hemopoietic and non-hemopoietic stem and non-stem cells of human cord blood leukoconcentrate in an autologous plasma, namely nucleated cells suspension of cord blood, suspended in an autologous plasma; white breedless female mice; hemopoietic organs (fetal liver) of Balb/ c mice embryos at different developmental stages and also industrial strains of cultures Streptomyces griseus 70, Streptomyces griseus 420, Streptomyces aureoverticillus 875, Streptomyces aureofaciens TB 633-FU, Bifidobacterium bifidum LBA-3, Lactobacillus bulgaricus 1Z 03501, Spirulina platensis, Saccharomyces vini, Saccharomyces cerevisiae L-1 starter, bacteriophage T4, E.coli K-12, higher fungi mycelium: Pleurotus ostreatus Don-103 (D-103), NK 35 - winter; Michigan - summer, shiitake fungi Lentinus edodes of IBK-55 strain, inoculated Hela cells , L929 and MdBk cultures . The research aim was the studying of the influence of hypothermic storage conditions, cryopreservation and freeze-drying in the objects with different organization level, carrying-out the investigation of the "Hemocord" preparation preventive effect in experimental animals infected by A/Victoria influenza virus. There have been used the following research methods: viability estimation of microorganisms by Koch's "plate" method; determination of protein spectrum by spectro-photometrical method; cells supravital staining; determination of antibiotic activity by biological method; determination of cell types' population quantity by monoclonal antibodies; determination of hemopoietic cells preservation on the inclusion into their nuclear material of fluorescent probe - propidium iodide with flow cytometer; virusological research methods of hemoglutination reaction (HGR) and inhibition reaction of hemoglutination (IRHG); cytochemical and histological methods; morphological and morphometrical methods; statistical processing of the findings by Student's-Fisher method. Programmable freezers of bioobjects UOP-6, ZМ 1.00.00.00 and UKB-1 (Ukraine) ; freeze-drying device UZV-2; fluorescent microscope "CIEF"(Belgium), light biological microscope "BIOLAM P2" (LOMO, RF), flow cytometer "FACS Calibur" (BD, USA), laser scanning confocal microscope LSCM (ZEISS, Germany), spectrophotometer "Pye Unicam SP8000" (England), photoelectrocalorimeter FEK-56М (Russia), рН -meter "pH-150 МК" (Russia), centrifuge ОS-BМ (Russia), termostat chamber with shakers (Ukraine) have been used for carrying-out researches. There has been shown the effectiveness of single intranasal injection of "Hemocord" preparation and its components before influenza virus A/ Victoria infection, providing antiviral protection for animals during 6 months. There has been shown the development of antiviral immunity enabling the activation of antiviral protection. There have been determined the optimal parameters of cryopreservation process and hypothermic storage of biological objects of different organisation levels. There has been developed the sublimated process technology of freeze-drying streptomyces vegetative culture, bifidobacterium, wine yeast and also complex probiotic preparation. For the first time it has been shown that "Hemocord " preparation and its fractions have a manifested ability to prevent a viral infection. For the first time there has been developed the protective medium and freeze -drying methods of industrial microorganism strains. There has been stipulated an application expediency of "Hemocord" preparation for prevention viral infections during the experiment, determined by activation of immune processes in an organism and its increasing its resistance to respiratory viral infections. There have been stipulated the optimal parameters of freeze-drying process for long-term industrial collections of microorganism strains' storage. Researches results are applied in medicine, veterinary, food, baking, microbiology industries, wine production and agriculture.
