This suggests that individual cancer cell types differ in the level of sensitivity to arginine starvation and that the mechanisms that underline these differences need to be further investigated. Obtained data revealed that among four studied amino acids deprivation for semiessential amino acid arginine and essential amino acid methionine had stronger inhibitory impact on cell growth and viability in comparison with deprivation for such essential amino acids as leucine or lysine. It was shown that the lack of arginine metabolites, polyamines and agmatine, was not the key reason of cell growth inhibition in arginine-free medium and did not determined the differences in the level of cancer cells sensitivity to arginine withdrawal. We also revealed that supplementation of arginine-free culture medium with nitric oxide donor enhanced the cytotoxic properties of arginine withdrawal against human malignant suspension cells.
