There were chosen conditions for direct and indirect Gentiana L. species organogenesis; obtained and rooted regenerated plants. It was established that the chosen cultivation conditions provide both growth of callus tissues and isolated root cultures of Gentiana species, as well as synthesis of biologically active substances (BAS) such as flavonoids and xanthones in them. In most cases the amount of BAS in dry biomass of culture in vitro exceeded or was close to that in intact plants roots. For the first time Gentiana species genomic variability was investigated by RAPD-and ISSR-PCR methods. It was found that Gentiana species variability did not transcend intraspecific polymorphism. The level of genetic changes was the lowest in regenerated plants obtained by direct organogenesis and the highest - in long-term cultivated non-morphogenic calli and isolated root cultures. It was shown that the developed technologies of obtaining and growing Gentiana.species morphogenic and non-morphogenic in vitro cultures at the background of low level genome variability provide BAS synthesis in them, the content of which in most cases exceeds or approximates to that in the intact plants roots. This creates the prospects of using the technologies developed, firstly, to obtain alternative sources of raw materials for pharmaceutics, secondly, to preserve the gene pool of these rare species.
