At executing of tasks of the first stage of the project the methodical approach, which allows to watch a course of reaction of disclosing thiazole cycle of thiamine in water solutions under the conditions close to the physiologic, has been developed. The action of some biologically active substances on reaction thiolozation of thiamine and ThDP is investigated with use of this method. It is found that among cations the most active is ion Zn2 + which activates speed of reaction for ThDP almost three times. Taking into account this effect the new sensitive express method of determination of ThDP contents in pharmaceutical preparations is offered. In experiences in vitro it is shown that under modelling conditions a peroxide of hydrogen, endogenic irradiation and menadion slow down the reaction of interaction of thiamine and ThDP with a Ellman reagent, that testifies to presence of direct action of these factors on thiamine molecule. In blood preparations thiamine brakes the formation of reactive oxygen species (ROS), induced by a peroxide of hydrogen or menadion that is accompanied by formation of the oxidized forms of thiamine, in particular, thiohrom. Increase of ROS level in blood of the rats, caused by introduction in vivo doxorubicin, is accompanied by decrease in blood level of SH-groups (reduced glutathione), and also formation of disulfide ThDP. The level of disulfide of ThDP in blood directly correlates with ROS level under these conditions.
