Investigation of the state of epithelium and biology of basal cells and expression of markers of the epithelial stem /progenitor cells p63, CK5/14 ; biomarkers p53, Bcl-2, Ki-67 in epithelial layers of normal oral mucosa, of precancerous lesions , in tumor-distant mucosa and tumors in patiens with OSCC and epidemiological risk. Metods . The biopsy specimens of oral mucosa and tumors were classified according to the UICC criteria. Tissue sections were immunohistochemically stained using monoclonal antibodies: for p63 (clone А4А), for C5/C14 (clone LL002 ) for p53 (clone DO-7), for Кі-67 (clone МІВ-1) ,for Всl-2 (clone 124), (Denmark) and counted according to epithelial layers as labeling index (LI, %). Results. Basal cells of normal epithelium of oral mucosa strongly expressed the marker proteins of epithelial stem /progenitor cells p63, CK5/14 as the predominant. In oral mucosa paties with epidemiological risk , precancerous lesions, in tumor-distant mucosa revaled the progression of histopathological phenotype (hyperplasia, dysplasia, carcinoma in situ, invasive carcinoma) and expression of proteins p63, CK5/14 in not characteristic suprabasal layers of epithelium of mucosa. The suprabasal expression of proteins p63, CK5/14, p53 correlated with grade of epithelial dysplasia and risk of oral cancer. The labeling index (LI, %) of p63, CK5/14, p53 cells were hight in invasive carcinomas . This data demonstrate the participate of cells that expression of markers of epithelial stem /progenitor cells in development of transformation phenotype. and biology of OSCCs. Conclusion.The immunohistochemicall labeling index of p63, CK5/14, p53 cells in suprabasal epithelial layers of oral mucosa can be used for differential diagnostics of grade of epithelial dysplasia and carcinoma in situ.
