Purpose - to study the severity of apoptosis in brain cells of rats of different ages within various periods after modelling of severe TBI , as well as an opportunity to influence the level of apoptosis by gene therapy aimed at induction of synthesis of apolipoprotein E3 isoform in the nervous tissue. Research Methods - We used the methods of experimental neurotraumatology (TBI modelling , intraventricular infusion of drugs by surgical implantation of brain cannula and osmotic pump ), molecular and genetic methods (molecular cloning, creation of expression constructions containing apoE3 cDNA gene , liposomal transfection , polymerase chain reaction) , morphological methods ( light optic and electron microscopy, morphometry, TUNEL method to determine cell death ). In the model of severe diffuse traumatic brain injury ( TBI) the impact of apoE3 gene therapy ,as well the processes of post-traumatic cell death in rats aged 8 and 24 months were analized. We found lower levels of apoptosis in rat brain at the day 10 after the injury , compared to day 5. Marked expression of different apoptotic processes in different areas of the brain after traumatic brain injury in rats of both age groups: highest in the cortex, slightly lower - in the hypothalamus , the lowest - in the hippocampus , which may reflect different intensity and timing of apoptotic processes in different areas of the brain. In all groups the severity of apoptosis was significantly higher in old rats than in adult animals. However, the degree of increase (relative to baseline) apoptotic index in adult rats was significantly higher than in the old animals. Data demonstrate that gene therapy produces some inhibitory effect on the process of cell death in the CA1 area of the hippocampus , reducing post-traumatic loss of neurons , while in adults the decrease is more pronounced than in older animals. It produces a positive effect on the brain as a whole, reducing the inflammatory and apoptotic processes and improving trophic after TBI . Area of application - experimental neurotraumatology.
