At the І stage by use of BLAST SEARCH of data base NCBI the primers design was developed to be used for PCR of exons of HFE1 gene; exons and introns of ITPA gene. Set of oligonucleotide primers were sythesized by solid phase phosphoamitide method and purified to be used for amplification by PCR of the sequences that are the components of the test system to be developed. Methods of mutation and polymorphism of HFE1 and ITPA genes analyses were developed and verified, the components of reactive mixture and temperature and time conditions were optimised, the test system components were produced. Analyses of allele polymorphism of HFE1 gene, a marker for hepatite C course, and ITPA gene, a diagnostic indicator of side defects development at hepatitis C patients in Ukrainian population after standard treatment, was conducted . Population frequency and spectrum of mutations and polymorphism in these genes were detected. Collection of DNA control samples with different 94С>А and IVS2+21A>C polymorphisms of ITPA gene and 845G>A mutation and 187 C>G polymorphism of HFE1 gene were created.
