Comparative isolation of MSCs from human umbilical cords using the enzymatic method and method of explants, and their cultivation in preparative amounts with preserved multipotent properties were performed. Individualized approach to the isolation method was developed based on the processing of 40 samples of umbilical cord. Cells from umbilical cord matrix received by these methods, had all the characteristics of mesenchymal stromal ( stem ) cells both in morphology and in the level of expression of surface markers, that decreased in passages ex vivo. The data obtained allow to consider MSC of 1-2 passages suitable for transplantation. The amount of MSCs received from from 10cm of umbilical cord was at least 5x107 , this number is enough for experiment of MSCs transplantation. Approaches for the enhancement of proliferative activity of MSCs received and period of high surface markers expression preservation were developed by MSCs culturing in nitrogen and argon.
