In particular, the impact of arginine deficiency on signaling pathways that regulate cell division, protein synthesis, apoptosis and cell adhesion were monitored. Two human colorectal carcinomas cell lines, HCT-116 and HT29 that differ in the sensitivity to arginine deprivation were used as models. It was shown that activation of GCN2/eIF2а, MAPK/ERK та mTOR signaling cascades, but not regulation of apoptosis initiation, defined cancer cell sensibility to the lack of arginine between different cell lines as well as between monolayer and spheroid culture of the same cell line. We revealed that arginine limitation reduced the adhesion of human colorectal cancer cells, HCT-116 and HT29, in monolayer cell culture. It was also demonstrated that human cancer cells grown in a form spheroids were more resistant to an individual amino acid deprivation in comparison with their monolayer cultures. Our data pointed out that the spheroid culture of human cancer cells are a better model than monolayer culture for investigation of anticancer potential of metabolic therapeutic approaches based on single amino acid deprivation in human body.
