The aim - to investigate barley polymorphism of DNA loci associated with response to unfavorable temperature conditions for the development of components of marker system of choice of resistant genotypes with a particular combination of allelic variants of studied genes. Methods – laboratory techniques, analysis of data using computer programs. We used the isolation of DNA with CTAB, extraction of total RNA, synthesis of cDNA by reverse transcription, amplification of DNA by PCR (Terzic, Russia), real-time PCR (iCycler iQ5, Bio-Rad), electrophoresis of PCR products in agarose and polyacrylamide gels, a computer program GeneFISHER, GraphPad Prism5, Blast. The allelic characteristics of barley varieties PBGI and other originators were obtained for genes / loci associated with resistance to unfavorable temperature conditions, namely 51 barley varieties - for locus Dhn 5, 39 varieties - for loci Blt 4.9, Blt 14, pAF93, pAО 29, РА986, Соr14b, Wcs 120.1, Dhn 1, Dhn 2, Dhn 3, Dhn 4, Dhn 7, Dhn 9, Cbf 4B, Cbf 12, Hva1, gene-base microsatellite loci (EST-SSR) Hv_gene-SSR24 (Dhn4), Hv_gene-SSR13 (Cbf10B), Hv_gene-SSR21 (Cbf4A), which take a part in the formation of resistence to low temperature as well as for locus Dhn3, MGB318, Leа affecting the drought. The presence of polymorphisms in exon 1 of Dhn4 gene and exon 1 Dhn7 gene were identified in the investigated barley varieties that is one of the reasons that lead to an modification in sensitivity to low temperature. The polymorphism in the regulatory region of Dhn5 gene was identified by PCR analysis and potential influences of transcription factors on gene expression during stress. Differences in allelic state Hva1 (target gene for transcription factor CBF2) were detected at the molecular level in barley genotypes contradiction of resistant to low temperature. The allelic state of Vrn- and Ppd-Н1-loci for 20 varieties of winter barley and intermediate was identified. The perspective is the use of expression level genes, in particular Dhn5 as marker for resistance to low temperature. The polymorphisms of locus Hva1 can be useful in plant breeding practice to identify winter and intermediate or spring genotypes in total sample varieties.
