The purpose of research is to obtain biologically active natural phosphopeptides with maximum preservation of their structure and characteristics. Phosphoproteins of milk were selected as promising precursors of bioactive phosphopeptides. For the first time it is shown that homogeneous phosphoprotein fractions of milk (alphaS1-CN, alphaS2-CN, beta-CN and kappa-CN) in gram quantities for one stage of purification can be obtained using preparative variant of anodic electrophoretic system with homogeneous PAAG. With exclusion chromatography the homogeneous fraction beta-CN was obtained. Through a combination of differential precipitation in the presence of urea and specific proteolysis with a recombinant chymosin was obtained the mixture alphaS1-СN-ХР and alphaS2-СN-ХР, which provided the higher output of phosphopeptides among others mixtures. It was determined that specificity of proteolytic preparation, pH, temperature and the stage of proteolysis affect on the molecular mass and on the structure of casein phosphopeptides. Therefore, for providing the formation of natural bioactive casein phosphopeptides the conditions of proteolysis of phosphoprotein substrates, which are implemented at physiological pH and temperature, were proposed. Specified conditions of natural casein phosphopeptides obtaining can be used for industrial obtaining them from the secondary raw materials, in particular from skim milk.
