Aim of the scientific investigations in the frames of the projects was elucidation of the dependence (at quantitative level) between cell ability to adhere on 2D solid surfaces and physical-chemical features of cell membranes, substrate and extracellular environment prior to and after cryopreservation. Methods of investigation: physical-mathematical analysis and numerical simulation, light and confocal microscopy, spectrophotometry, hydrodynamic studies, multi-step freezing. The shear stress values were found, which resulted in cell detaching, and their dependence on the duration of their adhesion and rate of spreading on a substrate; cell-cell adhesive interaction between erythrocytes and S. thermophilus lactobacteria was established to depend on medium composition and physical-chemical properties; the contribution of surface charge of cells and electrostatic interactions to cell-cell adhesion was assessed theoretically by erythrocyte surface potential, cation concentration near cell surface and Debye length in solutions with various physical-chemical characteristics; the developed physical-mathematical model was used to estimate the probability of specific adhesion appearance considering the Ca(2+) ion effect on cell electrostatic repulsion force and the inactivation of specific receptors resulting from their binding with the ions. The found regularities explain the contribution of cell electric characteristics and extracellular medium composition to adhesion processes. Development of the device for quantitative estimation of the forces of cell adhesion on solid surface provided the possibility to analyse quantitatively the adhesion process in dependence of cell state at various stages of cryopreservation. The found data could be used for development of the indices of cell surface structure preservation and, thus, to determine cell cryopreservation conditions being optimal with regard to adhesion abilities.
