They are expressed in both cell plasma membrane and mitochondria of eukaryotes. Neuroinflammation decreases the alfa 7 nAChR density in the brains of mice, is accompanied by accumulation of amyloid-beta (A-betta) and memory impairment. The aim of the present study was to reveal the effect of inflammation on the brain mitochondria. Inflammation was stimulated by either injections of bacterial lipopolysaccharide (LPS) or immunizations with alfa 7 nAChR extracellular domain (1-208). By means of various modifications of sandwich-ELISA assay, the decrease of alfa 7 nAChRs and accumulation of A-betta (1-40) and A-bretta (1-42) were found in mitochondria of immunized mice or those injected with LPS compared to control animals. Mitochondria of experimental mice started releasing cytochrome c at lower Ca2+ concentrations and were less sensitive to alfa 7 nAChR agonist PNU282987. It is concluded that inflammation alfa 7 nAChR expression in mitochondria, similarly to the plasma membrane, and makes mitochondria more susceptible to apoptosis induction.
