Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 The profile of tumor microRNAs associated with the aggressive clinical course of breast cancer was defined. Increased levels oncogenic miR-21 and lower levels of oncosuppressive miR-122, -200b were found in patients with stage III breast cancer compared with patients with stage I and II breast cancer (r = -0,59; r = 0,64 and r = 0, 56, respectively). Low rates of expression of oncosuppressive miR-20b and let-7 (r = -0,67 and r = -0,53) and high expression of oncogenic miR-10b, -21 and 149 (r = 0,78; r = 0,62 ; r = 0,54, respectively) were determined in tumors of patients with the presence of metastases in lymph nodes and correlated with adhesive properties of cancer cells (expression of E- and N-cadherin). The high degree of differentiation and low proliferative activity (Ki-67) of breast cancer correlated with inhibition of expression of oncogenic miR-10b expression and activation of oncosuppressive miR-200b and -320a (r = -0,52; r = 0,57 and r = 0,59 respectively). A distinguishing feature of the basal subtype of breast cancer were lower levels of miR-29c and miR-125b and increased levels of miR-155 and miR-199 in tumor cells. In luminal and Her2 / neu-positive breast tumors high levels of miR-205, -342 and -520a were established. Also, direct correlation of expression of miR-31 and inverse relationship levels of miR-155 and miR-199th on the presence of estrogen receptors in breast cancer cells were determined (r = 0,65; r = -0,53; r = 0, -69, respectively ). Downregulation of oncosuppressive miR-31, -122 and -133, as well as increased levels of oncogenic miR-27a, -155 and 182 in breast cancer tumor tissue were correlated with lower rates of disease-free survival and overall survival of patients. Based on a comprehensive assessment of molecular profile and miRNA profile of tumor cells new innovative biomarker panel monitor breast cancer was designed. Created algorithm of formation patients groups with high risk of tumor progression will allow a personalized correction of treatment regimen and improve the quality of life of cancer patients.
