Methods: In this paper have been involved in the cultivation of the adhesive and spheroid cell culture; immunocytochemistry (color of the tumor-associated markers: EGFR, ER, EpCAM, p53, Cks, Vim), cytology (study of different populations of multicellular tumor spheroid cells), flow cytometric (enzyme immunoassay - an ELISA, analysis of the proliferative activity of cells - MTT-test), molecular and biological, biochemical and statistical analysis methods. The aim of the study was to investigate the effect of cell-derived factor (interferon alpha and gamma factors of mesenchymal stem cells), and drugs (tamoxifen) to antigenic profile, clonogenic and metastatic potential of monolayer cell cultures and different subpopulations of cells of a multicellular tumor spheroid. The objectives of the study was cultured adherent breast cancer cells and generate one multicellular tumor spheroids. To determine the proliferative and morphological characteristics of tumor cells under the influence of the microenvironment factors and anticancer drugs in conditions 2D- and 3D-growth. As a result of different reactivity and established tumor cell viability in the monolayer and multicellular tumor spheroids. It is found that depending on the type of cell growth, expression of tumor-associated markers, proliferation and cell survival in the presence of humoral factors and different anti-cancer drugs. For example, in 2D culture observed uniform diffuse distribution of expression of the tumor markers (EGFR, ER, EpCAM, p53, Vim, Cks), in contrast to the cell aggregates in which the markers are expressed in the outer layers of the spheroids. In general, it was found that the factors that are produced by T cells have a pro-oncogenic effect on the tumor cell population as enhance proliferation, reducing adhesion, promote the expression of ER and EGFR, as well as stimulate the formation of multicellular spheroids. In turn, the factors on fibroblast cell division inhibit tumor cells and inhibit the formation of aggregates. Tamoxifen has cytotoxic, interferons alpha, gamma and humoral factors of mesenchymal stem cells - cytotoxic and estradiol - proproliferation effect in both cell cultures. The combined effect of tamoxifen with interferon ? deters migration ability of tumor cells. Metastasis cell potential increases by incubation with factors from the last T-lymphocytes and estradiol. Scientific and practical significance. It is expected that the results of research should be a necessary step to explore ways and mechanisms of action of the system of personalized therapy to stem tumor cells on the basis of multicellular tumor spheroids.
