Karazin popup.description2 The goal of the study consisted in the theoretical and experimental investigation of the mechanisms underlying the Doppler spectral response of soft tissues and development of the novel approaches to fluorescent monitoring of the biological systems of different levels of complexity. The objects of the study: soft tissues and model membrane systems of various composition, oligomers and fibrils of lysozyme, fibrils of apolipoprotein A-I mutants, hemoglobin, new fluorescent dyes of various classes. Methods of investigation: ultrasound scanning, fluorescence spectroscopy, spectrophotometry, quantum-chemical calculations, computer modeling, nonlinear optimization, mathematical statistics. The continual model of ultrasound waves scattering on the inhomogeneity of density s and compressibility, allowing for the acceleration of blood flows has been suggested. An original algorithm for calculating the ultrasonic attenuation coefficient has been developed. This algorithm is based on the amplitude analysis of ultrasonic response signals from different depths of tissue probing by the ultrasonic pulses and application of an iterative procedure which greatly improves the accuracy of calculations. Using the fluorescence spectroscopy technique the novel information on the interaction of the novel benzanthrone and ICT probes with the model lipid membranes has been obtained and the membrane-modifying effects of the fibrillar aggregates, oligomers and monomers of lysozyme and 1-83 fragment of apolipoprotein A-I have been studied. The oligomeric intermediates of amyloid fibrils have been shown to display the highest membranotropic activity. The enhancement of the membrane-modifying effects of the lysozyme prefibrillar aggregates in the presence of hemoglobin has been revealed. The results obtained are of importance for the improvement of the quality of ultrasound diagnostic technologies, as well as for the development of the nolvel strategies for diagnostic and treatment of amyloid pathologies.
