The goal of the study is to work out the biocenotic and genetic principles of the introduction of nature-approximate forestry in Ukraine, to find out the molecular mechanisms of the immune response of Scots pine, the main forest-forming breed of Ukraine, to the action of phytopathogens and to determine the prospects for using pine resistance genes as markers for estimating tree genotypes for resistance to diseases. Methods of research - forestry, forest assesment, microbiological, molecular biological, genetic engineering, bioinformatic, statistical. The biocenotic concept of introduction of nature-approximate forestry in Ukraine was presented in the paper. The processes of natural renewal of wood species, methods and mechanisms of reforming the same-age tree stands into different age-old forests were covered. The differential expression profiles of the defensin genes in pine seedling tissues on response to pathogen with different life-style were revealed. The molecular cloning of the Scots pine defensin gene 1 was performed and its intron-exon structure was established. The sequence of this gene was deposited in the GenBank database under number KH 690107. The molecular genetic methods of diagnosing pathogens of root rot, fusariosis and bacterial dropsy of pine trees have been developed, that allow identification of the pathogens on the early stages of infection.
