Object of research: multicellular tumor spheroids of the MCF-7 line, enriched with stem tumor cells (OBPS), human mesenchymal stem cells, interferon alpha. Research methods: the methods of cultivating adhesive and spheroid cell culture were involved; immunohistochemical method, method of morphometry (calculation of volumes of cell aggregates), analysis of cell proliferative activity (MTT-test) and statistical methods of analysis. The aim of the study was to analyze the influence of factors of cellular origin (interferon alpha and factors from mesenchymal stem cells) on the antigenic profile, clonogenic and metastatic potential of subpopulations of stem tumor cells of a multicellular tumor spheroid. Objectives of the study: to work out the method of enrichment of multicellular tumor spheroids (OBPS) from MCF-7 cell line with stem tumor cells (SEC). Prove the presence of SEC in OBBS. Determine the sensitivity of OBPTs to the action of cellular microenvironment factors (interferon alpha and factors from mesenchymal stem cells) and anticancer drugs (Cisplatin, Methotrexate, Doxorubicin). As a result of the work, an increase in the expression of markers of SEC breast cancer (CD24, CD44, CD133, bmi-1) was detected after enrichment of the SPS BPS culture. OBBS culture is less sensitive to the cytotoxic effect of anticancer drugs (Cisplatin, Methotrexate and Doxorubicin), compared to BPS culture, which was cultivated under standard conditions in a complete nutrient medium without adding growth factors. IFN-? dose-dependently contributes to the breakdown of OBPP and the formation of a large number of small cell aggregates and individual cells of the MCF-7 line. K-medium from MSC stimulates the formation of larger OBBPS and increases the survival of tumor cells MCF-7, compared with the control samples. Scientific and practical significance. It is expected that the use of OBPS in the SEC study can be used to create and test systemic antitumor therapy aimed at SEC.
