The goal is to evaluate the possibilities of using CNTs and their derivatives, namely BHT-Doc, in antitumor therapy. Research methods - histological, cytomorphological, microscopic, hematology. As a result of the work, a carbon compound was obtained, functionalization of an antitumor drug and a fluorescent label. In a monolayer culture, oxidized CNTs and BHT-Doc have a moderate cytotoxic effect at low concentrations (12.5 ?g/ml). With an increase in concentration to 25-50-100 ?g / ml, the viability of tumor cells decreases dose-dependently. With an increase in the concentration of CNTs to 200 ?g / ml, the viability of NT29 decreased to 39.2% for BHTox and 50% for BHT-Doc. In a spheroidal culture of tumor cells, it was shown that oxidized CNTs can stimulate the formation of BPS by almost 10 times. BHT-Doc stimulate the formation of BPS by 2.83 times. In most experiments, BHT-Doc has no cytotoxic effect on tumor cells in both 2D and 3D culture, which could be compared with the effect of a single doxorubicin. The simultaneous use of low concentrations of trypsin and BHT-Doc significantly increases the cytotoxic effect of CNTs and Doxorubicin compared to using these substances separately. When studying the effects of BHTox and BHT-Doc in vivo, the greatest effect was found on hepatic enzymes. In hematological studies, signs of hematological toxicity of CNT-Doc and pure Doxurubicin were revealed. What finds expression in dose-dependent myelosuppression, leukopenia and neutropenia (neutropenia). With the introduction of CNT-Doc, pathological tissue remodeling of the liver is determined. Thus, based on the results obtained, we assume that the carbon-protein construct (BHT-Doc) is able to carry the drug in the cell microenvironment but does not cause significant cytotoxic effects. In the presence of peptidase (trypsin), doxorubicin is released and its cytotoxic effect increases. The indicated effect was observed in experiments both in vitro and in vivo.
