The conducted studies have shown possible ways to improve the enzyme in order to modify the spectrum of indole alkaloids in plants by the methods of metabolic engineering. The new pigments – сeloscristatyn and its isomers were discovered in the Celosia cristata L. callus culture. This callus culture is the only one known source of сeloscristatyn and its derivatives. The ability of callus culture of Celosia cristata L. to produce significant amounts of dopamine has been confirmed as well. Amount of dopamine in the callus culture is twice as large as in inflorescences of Celosia cristata L. collected in nature. Biotechnologies of the accumulation of recombinant compounds in plants as a result of the expression of heterologous genes are presented in the work both for genetically modified plants and for transient expression of heterologous genes. The method of Agrobacterium-mediated transient expression of recombinant proteins by the plant system in vitro with the use of a modular system of vectors based on elements of viral replicons has been developed. In the framework of this project transgenic plants of different species were obtained. For the genetic transformation of plants vectors containing the genes of secretory antigens ESAT6 and Ag85b that stimulate the immune response to tuberculosis were used. In this work transgenic potato plants which accumulate the amino acid proline were also obtained. The biotechnology for collicin production in plant systems has been developed as well. During the reporting period a collection of in vitro rare species of the genus Crambe L. was created and protocols of microcollonal reproduction that could be used for further return to natural growth sites of these plants were developed.
