We have performed in vitro comparison and optimization of ratios for different bone morphogenetic proteins (BMP) while aiming to improve their osteoinductive properties. We have found that BMP preparations generated as a result of cotransfection of 85% of optimized BMP2 and 15% BMP7 plasmids are most efficient inducers of early osteogenesis of mesenchymal stem cells (MSC). We also found that Secreted Phosphoprotein 24, Spp24 is a promicing modulator of BMP osteogenic effects and elucidated a molecular mechanism mediating a positive cooperation of BMP and SPP24 effects in osteogenesis. Polyuretan osteoplastic materials were generated as a cooperative effort with Gdansk Polytechnical Univarsity, Gdansk, Poland. We performed the optimization in vitro of conditions for incorporation of mesenchymal precursor cells into polyuretan osteoinductive matrices with incorporated BMP.
