The aim of the study is to analyze the changes induced by endothelial activation, stress and physical activity; to investigate the possibility of their correction with antioxidants; to introduce the experimental study of pulmonary arterial hypertension (PAH), to evaluate changes in adaptive potential under the influence of the oxidative stress. Methods - analysis of the publications; laboratory methods: biochemical blood analysis, enzyme immunoassays; physical exercising tests; statistical analysis. The results of the research has revealed that the simultaneous administration of PDE5 inhibitor sildenafil and antioxidants of food origin (quercitin) demonstrates the augmentation of the sildenafil induced vasorelaxantion. Although modern specific treatment improves the clinical course and prognosis of PAH, it is not possible to control sufficiently the mortal progression of the disease. According to the emerging evidences an initial combined PAH-specific treatment aimed on multiple pathogenetic mechanisms of the disease demonstrates the benefits for the prognosis. Understanding the important role of the right ventricular insufficiency (RVI) in the course of PAH stimulates the investigations for the new drugs designed to limit the progression of RVI. The results of the studies showed that vasorelaxant action of PDE5 inhibitor sildenafil can be potentiated by the simultaneous administration of quercitin, an antioxidant of food origin.
