Subject of study: Effect of exogenous polyamines, disaccharide trehalose and arginine deficiency on proteolytic autophagic degradation of human alpha-synuclein in model biosystems of H. polymorpha and D. melanogaster. The purpose of the research is to study the molecular mechanisms of degradation of abnormal forms of human alpha-synuclein in model organisms of H. polymorpha and D. melanogaster at introduction of exogenous polyamines and trehalose, as well as under arginine deficiency. Objective: analysis of effect of arginine deficiency (for H. polymorpha and D. melanogaster) and Nitrogen source deficiency (for H. polymorpha) on the degradation of abnormal (oligomeric, fibrillar) forms of human alpha-synuclein in model organisms; study of protective properties of agmatine (as an ROS scavenger) in cells of H. polymorpha model strains and dopaminergic neurons of transgenic D. melanogaster lines; investigation of role of spermidine and trehalose in the induction of autophagic degradation of human alpha-synuclein in the cells of model organisms. Performing these tasks will further develop new therapeutic approaches for the creation in human body of conditions for the effective autophagic degradation of damaged proteins and organelles using low molecular weight compounds of natural origin (such as polyamines or trehalose).
