The aim of the work is to develop a system for the accelerated creation of breeding material for bread winter and spring wheat, spring barley and winter triticale based on haploid biotechnologies, cell selection and molecular genetic markers. Research methods – tissue and organ culture in vitro, in vitro selection, PCR analysis, laboratory, field, statistical. Results – the individual elements of biotechnology were developed and improved for creating initial material for breeding cereal crops; new breeding material was obtained and stabilized. In a number of bread winter wheat varieties different types of wheat-rye translocations were found: 1AL.1RS and 1BL.1RS. The level of genetic diversity of winter wheat varieties was assessed using ISSR markers; these markers will be used in further breeding programs. The methods for obtaining dihaploid lines of cereals (winter and spring wheat and, spring barley, winter triticale) were improved using anther culture, isolated microspore culture and distant hybridization (selective chromosome elimination). The regenerated plants of bread winter and spring wheat, spring barley and winter triticale with complex tolerance to salinity and water deficit were obtained by direct and step cell selection methods. The cytological instability of regenerated plants obtained by cell selection on the ploidy level was found. The level of polymorphism of DNA regions flanked by inverted LTR repeats of retrotransposons in regenerated cereal plants obtained by cell selection for tolerance to osmotic stress was analyzed by IRAP analysis, and polymorphic loci were identified. The regenerated plants of bread winter and spring wheat and winter triticale, tolerant to culture filtrate Fusarium graminearum Schwabe were obtained by direct cell selection.
