Object of research: regulation of alcoholic fermentation of xylose in thermotolerant yeast Ogataea polymorpha. Aim of the study: to improve the yeast producers of ethanol by elucidating the mechanisms of regulation of alcoholic fermentation and the use of metabolic engineering. The purpose of the stage: to study the genetic and biochemical aspects of the regulation of alcoholic fermentation in yeast and the possibility of increasing ethanol production as a result of activation or inhibition of the expression of transcriptional activators or repressors Research methods: genetic engineering, microbiological, biochemical. Derivatives with deletion and enhanced expression of the HAP4 gene were constructed in xylose-fermenting strain of S. cerevisiae GS010. The deletion of HAP4 leads to an increase in the accumulation of ethanol from xylose, the mutant produces 10.4 g / l of ethanol Increased HAP4 expression resulted in decreased ethanol production from xylose and glucose. The deletion of the TUP1 gene causes a decrease in the fermentation of xylose to ethanol. The tup1Δ strain was characterized by impaired growth and the ethanol production on glucose medium. The deletion of the TUP1 gene in the wild-type strain of O. polymorpha causes an increase in ethanol production from xylose and a deterioration in the ability to ferment glucose. The deletion strains hap4AΔ and hap4AΔ hap4BΔ are characterized by increased ethanol production from xylose compared to the wild-type strain. Strains with enhanced expression of TUP1 or HAP4A genes are characterized by reduced production of ethanol from xylose. Increased expression of the HAP4B gene under the control of a promoter of the GAP1 gene or a native promoter did not have a positive effect on ethanol production from xylose.
