The object of the study - the process of obtaining a highly active virus-safe drug of coagulation factor VIII using affinity chromatography on silica sorbents. The aim of the work - to develop a method of obtaining a highly active virus-safe drug of coagulation factor VIII with the use of newly synthesized biospecific silica sorbents with immobilized active dyes and to study the properties of the obtained drugs. Research methods: chemical synthesis of sorbents; fractionation; dialysis; gel and ultradiafiltration; spectrophotometric; electrophoretic; affinity and ion exchange chromatography; coagulological and methods using hydrolysis of specific chromogenic substrates; antiviral treatment; enzyme-linked immunosorbent assays; statistical. The following scientific and practical results were obtained: from fresh-frozen plasma using the stages of viral inactivation, pre-fractionation and methods of negative affinity sorption created a scheme for obtaining purified FVIII; no residues of fibrinogen, thrombin, FIX, traces of virus-inactivating substances were detected in the drug; the drug is stable at room temperature 6 h, at + (4 ÷ 8) ° C - 24 h, in the frozen state (-30) ° C for up to 3 months, after lyophilization - 2 years. Degree of implementation: acts of implementation, reports at international scientific forums, publications in the scientific literature. Field of application: blood centers, chemical and pharmaceutical companies, research organizations. The implementation of research results will improve the technology of obtaining diagnostic and therapeutic drugs of factor VIII blood clotting from donor plasma. Were published: 14 scientific papers, including 3 articles, 10 abstracts, 5 of them abroad, 1 newsletter. The results are presented at 12 international forums.
